A MALDI-TOF MS database for fast identification of Vibrio spp. potentially pathogenic to marine mollusks ArchiMer

In mollusk aquaculture, a large number of Vibrio species are considered major pathogens. Conventional methods based on DNA amplification and sequencing used to accurately identify Vibrio species are unsuitable for monitoring programs because they are time-consuming and expensive. The aim of this study was, therefore, to develop the MALDI-TOF MS method in order to establish a rapid identification technique for a large panel of Vibrio species. Adherence assays and Slime production of Vibrio alginolyticus and Vibrio parahaemolyticus BJM

In this study we investigated the phenotypic slime production of Vibrio alginolyticus and Vibrio parahaemolyticus strains, food-borne pathogens, using a Congo red agar plate assay. Furthermore, we studied their ability to adhere to abiotic surfaces and Vero cells line. Our results showed that only V. alginolyticus ATCC 17749 was a slime-producer developing almost black colonies on Congo red agar plate. Adherence to glace tube showed that all V. alginolyticus strains were more adherent than V. parahaemolyticus. Only V. alginolyticus ATCC 17749 was found to be able to form biofilm on polystyrene microplate wells (OD570 = 0.532). Bacterial diseases in marine bivalves ArchiMer

Bivalve aquaculture is seriously affected by many bacterial pathogens that cause high losses in hatcheries as well as in natural beds. A number of Vibrio species, but also members of the genera Nocardia and Roseovarius, are considered important pathogens in aquaculture. The present work provides an updated overview of main diseases and implicated bacterial species affecting bivalves. This review focuses on aetiological agents, their diversity and virulence factors, the diagnostic methods available as well as information on the dynamics of the host-parasite relationship.

Bacterial problems associated with scallop Pecten maximus larval culture ArchiMer

Scallop Pecten maximus larvae have been cultured at the Argenton and Tinduff (Brittany, France) hatcheries with antibiotic treatment (chloramphenicol at 8 ppm) for 15 yr. Without treatment, outbreak of disease has normally occurred between Day 12 and Day 19 or sometimes earlier. A bacteriological study of larvae reared with and without antibiotic was performed over a 4 yr period. Among the collected strains, 2 clusters (C and F) of vibrios were present at high densities only in larvae cultured without treatment. Marennine, the water-soluble blue pigment produced by the marine diatom Haslea ostrearia, is known to display antibacterial activities. Previous studies have demonstrated a prophylactic effect of marennine on bivalve larvae challenged with a pathogenic Vibrio splendidus, suggesting that the blue Haslea is a good candidate for applications in aquaculture as a source of a natural antimicrobial agent. In French Polynesia, black pearl cultivation now takes place on about 30 of the 118 atolls or islands. This activity has largely replaced copra, coconut oil and fishing as a source of income. Around 1,000 pearl farms, generally small ones of less than 5 ha, are registered. The pearl farms are mainly owned by families. In 1998, about 7,000 persons were employed in theentire pearl industry, from production to retailing. In value, this activity represents the first exportation of French Polynesian products. From 1988 to date, exports increased exponentially before leveling. Starting with less than two kilograms in 1972, exports have now stabilized at Il tons of "black" pearls.

Water and sediment samples monthly collected from Arcachon Bay (Gironde, France) were examined during three years for the presence of marine vibrios. V. cholerae serotype 01 was never detected. On the other hand, the Eyre River and mouth contain non-agglutinating V. cholerae (127 strains) principally in water. The low temperatures slow their development down without stopping it completely. V. parahaemolyticus (257 strains) present in the same areas is particularly isolated from water when the aquatic medium temperature rises to 15 degree C. Understanding the ecological and evolutionary dynamics of infectious agents is important for diagnosing, predicting and preventing diseases in farmed and wild species. This project was aimed at studying the evolution of V. crassostreae virulence in relation to the oyster as a host and to the phages as predators.
